Quantitative Electron Microscopic Autoradiography

چکیده
Quantitative electron microscopic autoradiography was used for comparing the binding of labeled somatostatin-14 (S-14) and somatostatin-28 (S-28) to islet cells. Monolayer cultures of rat islet cells were incubated with ['251-Tyr"jS-14 (S-14) or [125I-Leu8, D-Trp22, TyreJS-28 (S-281) in the presence or absence of excess unlabeled peptides. Autoradiographic grains (ARG) associated with individual islet cells were identified and expressed as the mean number per B, A, and D cells. Specific ARG associated with S-14 were found over B and A cells. S-281-related specific ARG were concentrated over B, A, as well as D cells. The highest density of S-14§ labeling occurred over A cells, which under conditions of maximum labeling (370C for 60 min) contained five times as many ARG as did B cells. By contrast, under the same incubation conditions, the labeling density with S-281 was maximal over B cells, which contained four and five times as many grains as A and D cells, respectively. These observations show preferential association of S-14 with the A cell and S-281 with the B cell. They provide strong evidence for the existence of separate binding sites for S-14 and S-281 on A and B cells, respectively, which presumably mediate the previously reported glucagon selective inhibitory effect of S-14 and the insulin-selective action of S-28.

Direct demonstration of insulin receptor internalization. A quantitative electron microscopic study of covalently bound 125I-photoreactive insulin incubated with isolated hepatocytes.
When 125I-insulin is incubated with isolated rodent hepatocytes at 37 degrees C, the ligand initially binds to the plasma membrane of the cell and is subsequently internalized by adsorptive endocytosis. To confirm directly that the insulin receptor is internalized with the ligand, we covalently linked photoreactive 125I-N sigma B29 (azidobenzoyl) insulin to its specific hepatocyte receptor and ...

The retention and loss of PH]s-hydroxytryptamine (5-HT) from rabbit platelets during fixation was studied using liquid scintillation counting and quantitative electron-microscopic autoradiography. The results were at variance with previously reported data on human platelets. Following treatment of the platelets with either 2 0 % formaldehyde or 2 5 % glutaraldehyde for 2 min or 1 h it was found...

The sites of the incorporation of labeled cystine into keratinizing structures were studied in electron microscopic autoradiographs. The tracer used was cystine labeled with S(35) emitting long-range ionizing particles. During exposure for 1 to 2 months, according to our method of electron microscopic autoradiography, emulsion-coated specimens were exposed to a static magnetic field which appea...

Electron microscopic autoradiography with [3H]histidine, [3H]cystine, I13H]arginine, and [3H]proline was used to study protein synthesis in keratohyaline granules of newborn rats. All 3H-amino acids were incorporated into proteins in the granular cells, and the radioactive proteins appeared in the keratohyaline granules. However, the amount of radioactivity associated with the granules and the ...
متن کامل125I-insulin binding to cultured human lymphocytes. Initial localization and fate of hormone determined by quantitative electron microscopic autoradiography.
Morphologic and biochemical studies indicate that the initial action of insulin is binding to a cell surface receptor. Whether further translocation of the hormone, or a product of the hormone, occurs is unclear and has not been investigated by direct means. To determine the fate of 125I-insulin bound to its receptor, we have examined the distribution of radioactivity by quantitative electron m...
